The temporary medical testing centre at the unused Sainsbury's store near the Riverside Stadium will be relocating into the town centre.
As a result the car park opposite the Royal Mail sorting office has been closed to allow the site to be prepared for the move.
People visiting the town centre are advised that alternative long stay parking is available at Captain Cook Square multi-storey, nearby on Cannon Park Way, Station Street and the Zetland multi-storey.
Cllr Ashley Waters, Middlesbrough Council's Executive Member for Regeneration, said: "We want to make everyone aware that the Cannon Park Road car park is to be repurposed as a coronavirus medical testing facility.
"This will no doubt cause inconvenience to some but clearly our fight against Covid-19 must come first.
"We are fortunate to have a testing facility in Middlesbrough and hopefully it moving to the town centre will make it even more accessible to more people who need it."
